Quinomycin A reduces cyst progression in polycystic kidney disease

چکیده
Polycystic kidney disease (PKD) is a genetic disorder characterized by aberrant renal epithelial cell proliferation and formation progressive growth of numerous fluid-filled cysts within the kidneys. Previously, we showed that there elevated Notch signaling compared to normal cells contributes cystic cells. Quinomycin A, bis-intercalator peptide, has previously been shown target pathway inhibit tumor in cancer. Here, show A decreased cyst human ADPKD cultured 3D collagen gel. Treatment with reduced weight body ratio area fibrosis Pkd1RC/RC; Pkd2+/− mice, an orthologous PKD mouse model. This was accompanied expression proteins, RBPjk HeyL kidneys mice. treatments also normalized cilia length derived from collecting ducts. first study demonstrate effectively inhibits progression suggests potential therapeutic value for patients.

Pyrrolidine dithiocarbamate reduces the progression of total kidney volume and cyst enlargement in experimental polycystic kidney disease
Heterocyclic dithiocarbamates have anti-inflammatory and anti-proliferative effects in rodent models of chronic kidney disease. In this study, we tested the hypothesis that pyrrolidine dithiocarbamate (PDTC) reduces the progression of polycystic kidney disease (PKD). Male Lewis polycystic kidney (LPK) rats (an ortholog of Nek8/NPHP9) received intraperitoneal injections of either saline vehicle ...

2181 protect against lung injury in this clinical setting, but it seems unlikely. After almost 40 years of searching unsuccessfully for effective strategies to prevent broncho-pulmonary dysplasia, it would be a welcome surprise if a simple pharmacologic intervention proved to reduce its incidence. The findings of Schmidt et al. suggest the possibility that caffeine may have this effect, at leas...
متن کاملVolume progression in polycystic kidney disease.
BACKGROUND Autosomal dominant polycystic kidney disease (ADPKD) is characterized by progressive enlargement of cyst-filled kidneys. METHODS In a three-year study, we measured the rates of change in total kidney volume, total cyst volume, and iothalamate clearance in patients with ADPKD. Of a total of 241 patients, in 232 patients without azotemia who were 15 to 46 years old at baseline we use...
متن کاملCaspase inhibition reduces tubular apoptosis and proliferation and slows disease progression in polycystic kidney disease.
We have previously demonstrated an increase in proapoptotic caspase-3 in the kidney of Han:SPRD rats with polycystic kidney disease (PKD). The aim of the present study was to determine the effect of caspase inhibition on tubular cell apoptosis and proliferation, cyst formation, and renal failure in the Han:SPRD rat model of PKD. Heterozygous (Cy/+) and littermate control (+/+) male rats were we...
متن کاملMacrophages promote polycystic kidney disease progression
Renal M2-like macrophages have critical roles in tissue repair, stimulating tubule cell proliferation and, if they remain, fibrosis. M2-like macrophages have also been implicated in promoting cyst expansion in mouse models of autosomal dominant polycystic kidney disease (ADPKD). While renal macrophages have been documented in human ADPKD, there are no studies in autosomal recessive polycystic k...
